Vical Incorporated announces results of herpes vaccine trial

vical-incorporated-logo-300x106.jpgVical Incorporated, a San Diego-based gene-based treatment and vaccine company, announced the results on Monday of a study testing its Vaxfectin-formulated plasmid DNA herpes vaccine on guinea pigs.

The vaccine candidate was meant to protect against herpes simplex virus type two, the more serious of the two serotypes of herpes simplex. Approximately 25 percent of the U.S. population is seropositive for HSV-2 by the age of 40, Seeking Alpha reports.

The company tested multiple variants of the vaccine and found that Vaxfectin-gD2/UL46/UL47 was the most successful in reducing latent viral load of herpes simplex. The variant was also able to reduce both recurrent disease and viral shedding in the genital tract.

“The observation that therapeutic immunization with Vaxfectin-gD2/UL46/UL47 significantly reduced the frequency of virus shedding has public health implications given that this is thought to be a major source of transmission,” the authors of the study said, according to Seeking Alpha.

The authors said that the success of the Vaxfectin variant strongly suggests that it warrants development as a therapeutic vaccine and should be moved into clinical trials. The company anticipates that the HSV-2 vaccine could enter clinical trials in the second half of 2013.

The study says that while immunization reduced the potential for viral transmission by reducing the frequency of genital tract shedding, the potential for infection by exposure during a shedding event is not lessened.

There is currently no cure for genital herpes, Seeking Alpha reports.

Viral is developing the vaccine candidate under a grant from the National Institute of Allergy and Infectious Diseases.

The study was published in the journal Vaccine.

In addition, 40 percent of the mice immunized with the combined gD2 and Vaxfectin injection had no detectable HSV-2 remaining in nerve cells known to harbor the virus. This finding is important because it relates to the tendency of HSV-2 to take up permanent residence in a group of spinal nerve cells called the dorsal root ganglia. When reactivated, the virus emerges from this group of cells to cause genital herpes. (HSV-1 reactivates more efficiently from the trigeminal ganglia to cause cold sores or an inflammation of the cornea.)

Well it did confirm that they are going into clinical trials this year. I think that's a good thing. I'm actually hoping to move to Seattle when my lease is up at the end of the year. Vegas is nice but it's too dry here for my taste. Partly, to see if I can take part in clinicals & partly b/c I think it would be nice to live near the ocean for a bit.

Gothic Beauty, Thank you! That is a wonderful idea. I am happy you have the resources to make such a move. If I didn't have a child I would look into doing the same thing. :) Keep up posted. This forum is sooo great. I tend to just read the positive stuff around here so I don't get all frazzled by negativity. Again, thank you. If I EVER get the chance at a clinical trial I am going to do it. It's time that this disease is stopped! I will never pass this to someone, I know the emotional toll it has took on me and my day to day living (ups and downs) I just wish everyone had morals and wouldn't deceive innocent people. Takes two to tango tho! Trust has now become a huge issue as well for me. Take care and good luck on your adventure:)

I have a child but I have contacted numerous places to see if I could participate using my regular dr. He is extremely interested in this stuff and he is awesome. He even admits I know alot more about this stuff then he does. I'm lucky to have a supportive dr. I thought about moving there too. But sometimes the university of Washington allows outsiders to participate in other states if the have a good dr willing to do all this stuff and go in and do swabs everyday and blood tests. I'm going to try to see if my dr can contact them to see if I can get into one of them. If not I will move. I refuse to just sit here and deal with this. I have Tk deficient hsv2 so anti virals don't work for me and my outbreak hasn't stopped since I got diagnosed two months ago. It's even my daughters bday tomorrow and our favorite thing is swimming and now I can't go. I'm lucky my outbreaks I guess are mild for a newbie but my whole life has changed and it effects my daughter as well so I'm not gonna sit here and just accept my date quite yet.

I'm not concerned about not being willing to participate. I just think it would more convienient to live close to the facility that does the most research/work on it than anywhere else in the country. What's Tk mean?

      You obviously can’t read. Why don’t you actually read the article, or the excerpt I posted? Here it is even simpler for you: prostaglandins were shown to enhance the spread of herpes virus in cell culture 26 27 and to inhibit interferon production 27 28 and were implicated in the reactivation of herpetic skin lesions.

      Hormones definitely affect HSV outbreaks according to research. I'm a male but on anti-estrogen medication in constant trouble.       It's getting pretty terrible now, I don't ever remember the last time I was feeling normal. I'm so sick of this constant flu-like feeling that makes me depressed and very unproductive. I have so many ideas and ambitions for life but I end up just doing the bare minimum due to constantly feeling like shit.  A week ago I tried stopping my anti-estrogen medication if that could have been the cause (it states "infection" in the listed, common side effects). Had my hopes up for a day or two but just got another outbreak this week that makes me want to sleep in bed all day. On top of this, now my breasts hurt because of increased E2 levels. 

      I guess I still have some hopes with new research drugs such as Crispr and Amenalief. I would order Amenalief from Japan immediately, but it costs more than my monthly rent.. I might have to start a business on top of my full time job so I can afford it. Better option than self-pity but extra stress will surely not help with the HSV2 ;)

      Probably the therapy with SADBE helps us to develop a partial immune system on HSV. If you use more SADBE you will develop more immune system on HSV. This is most likely a mechanism of action.
